This is only a quess- But Iam thinking the center tube is #2 and the B+ is the upper copper strip going thru this circled resistor then to pin 1 tube 2? unplug and check these connections and use your meter if it looks ok.

using meter verify the bottom of this resistor is going to pin 1 .. you can check with the working side - the same on bottom of board that goes to pin 6

Or better yet check and make sure resistor didn't go open and reflow solder at pin 1 and this resistor.
